I guessing a blood stain from the gist of the story. Probably a whole lot of blood, too. 
So that would be a 'yes' - a whole lot of yes.
Not sure if a blood stain is considered a protein stain for cleaning purposes. True the red color comes from hemaglobin, a protein that contains iron, but I think you clean blood differently than other protein (Monica Lewinski- style) stains.
Solution... Incinerate the shirt, and then sell your shovel and hacksaw to buy a new one.
But the shirt has sentimental value. And I will probably be needing the shovel and hacksaw.
